The cover glass is a WINDOW of your sample. Through the cover glass you observe the specimen. The quality, thickness, size – those are crucial propertis of it and directly influence your results.
The slide is a CARRIER for your sample. It is not so much crucial for fluorescence, more important for transmitted light. Anyway, it could be precise and nice, with useful features and high suitability.
