1 - HIGH PRECISION COVER GLASS
Be careful with cover glass selection. The uniformity of the thickness is extremely beneficial when we are talking about advanced or super-resolution microscopy. This precision in glass thickness defines the reproducibility of your results. With one range of glass thickness you have similar shape of PSF and thus you have uniform results also from deconvolution. This effect is crucial, actually.
2 - APPROPRIATE MOUNTING MEDIUM
This step – in which mounting media the specimen will be mounted – is the key decision. It depends on several requirements, anyway for highest possible resolution you always select the highest possible refractive index of the mounting. As close as possible to the glass refractive index. Sometimes, you have some special fluorophores or samples and this step is possible to optimize.
3 - EASY MOUNTING SPACER
And what is the mounting spacer? This small thing would have dramatic effect on the sample quality and vitality. As we understand the spacer, it is both-side sticky component with precisely defined thickness which is helpful for instant mounting and sealing, moreover and mainly – it prevents any pressure from cover glass and slide to the specimen and preserve it against structural collapse.
